Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hIGF-II (human IGF-II ). Anti-Human IGF-II specific antibody was purified by affinity chromatography employing immobilized hhIGF-II matrix.

This antibody stained formalin-fixed, paraffin-embedded sections of normal human placenta. The recommended concentration ranges from 0.5 µg/ml-5.0µg/ml. An HRP-labeled polymer detection system was used with DAB chromogen. Recommended antigen retrievals are heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer or enzyme induced antigen retrieval with Proteinase K. Optimal concentrations and conditions may vary.
To yield one-half maximal inhibition [ND50] of the biological activity of hIGF-II (10 ng/ml), a concentration of 3.1 - 4.6 µg/ml of this antibody is required.
Sandwich ELISA:
To detect hIGF-II by sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.5 - 2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with BioGems' Biotinylated Anti-Human IGF-II (60-187BT) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hIGF-II.
Western Blot:
To detect hIGF-II by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIGF-II is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
